三七复合有效成分对子宫内膜炎症细胞NF-κB p65及TNF-α、IL-1β变化的影响 被引量:9

摘要 目的:探讨三七复合有效成分在子宫内膜细胞炎症过程中对NF-κBp65表达和活性变化及其TNF-α、IL-1β含量变化的影响。方法:应用WB,ICC,EMSA方法分析子宫内膜细胞LPS炎症模型组、三七复合有效成分组、宫血宁制剂组及正常对照组NF-κBp65的表达情况以及其活性的动态变化;应用ELISA法测定TNF-α、IL-1β的含量。结果:子宫内膜细胞炎症过程中细胞核内出现NF-κBp65的高水平表达(P<0.01),TNF-α、IL-1β的含量随炎症发展而逐渐上升(P<0.01),三七复合有效成分能有效降低炎症因子TNF-α、IL-1β的含量,降低NF-κBp65的转录及蛋白表达量,抑制NF-κBp65的核因子DNA结合活性。结论:三七复合有效成分能促使炎症细胞转归,有效保护LPS对培养细胞的损伤,在长效持续性保护细胞形态完整性方面优于宫血宁制剂。 Objective: To investigate the change of nuclear factor κBp65 and TNF-α, IL-1β expression in the inflammatory model of human endometrial cell affected by the compound components from panax notoginseng. Methods: EMSA, ICC and WB were used to detect the activity of NFκBp65 , the ELISA were used to detect the concentration of TNF-α and IL-1β. Results: Nuclear translocation of NF-κBp65 appeared in the progression of infection model of human endometrial cell, the activity of NF-κBp65 were increased obviously in nuclear, and the concentration of TNF-α, IL-1β also increased. The compound components from panax notoginseng could inhibit the expression of TNF-α, IL-1β and NF-κBp65 , the transcription of NF-κBp65, and the DNA binding activity of NF-κBp65 . Conclusion: The compound components from panax notoginseng can reduce the intensity of inflammation and especially have predominance on protection of integrality of endometrial cell contrasted with gongxuening capsule.
