摘要
作者利用纯化的伤寒杆菌鞭毛抗原，制备出抗伤寒杆菌鞭毛抗原单克隆抗体。实验发现，四株单抗与伤寒杆菌鞭毛抗原和伤寒杆菌发生免疫反应，与甲、乙、丙副伤寒杆菌、大肠杆菌及部分沙门氏菌无交叉反应。选用其中２株单抗分别作为包被抗体和标记抗体，建立了检测伤寒杆菌鞭毛抗原的夹心ＥＬＩＳＡ。３０名正常人和４２例非伤寒发热待查患者血清标本经检测为阴性，１２例伤寒患者血清标本为阳性。本方法可直接用血清进行检测，具有早期应用、快速、简便和特异性强等特点。
Four monoclonal antibodies (MAbs) to Salmonella typhi flagellin were prepared and no immunological reaction was found between them and other enterobacteria strains including S. paratyphi A,B,C,E. coli,S. enteritidis,S. typhi murium and S. chlolerae suis. With two of them a double sandwich enzyme-linked immunosorbent assay (ELISA) was developed to detect S. typhi flagellin in serum samples. S. typhi flagellin was detected in all serum samples from 12 patients with positive blood culture for S. typhi and in 4 patients with negative blood culture, positive results were also obtained. Serum samples from 30 blood donors and 46 fever of unknown origin showed negative results. The ELISA for S. typhi flagellin is simple,quick and may have practical value for rapid diagnosis of typhoid fever.
