摘要
采用直接提取法对活性污泥总DNA进行提取,以化学机械法为基础,在细胞裂解步骤中,分别设计了化学法与超声法、高温法、反复冻融法、珠磨法及液氮研磨法相结合等方法,并通过DNA样品的浓度、纯度、提取率以及肠杆菌基因间重复共有序列扩增片段多态性等4个指标对活性污泥总DNA提取的不同细胞裂解方法进行比较,确定了反复冻融法为提取效率高、DNA纯度好且适用于PCR扩增的最佳DNA提取方法.
Directed-extraction method was used for DNA extraction from activated sludge. Based on the chemicallysis and Mechinablysis, different DNA extraction methods for activated sludge were evaluated. They are ultrasonicaUysis, high temperature extraction, freeze-thawing, head mill homogenization and liquid nitrogen method. The effects of them were compared based on the concentration, purity, efficiency of distillation of the recovered DNA, and length polymorphism of ERIC-PCR products.
