摘要
目的:观察亚低温对局灶性脑缺血大鼠神经细胞凋亡及Bcl-2蛋白表达影响,探讨亚低温的脑保护作用。方法:选择健康雄性大鼠,随即分为假手术组、脑缺血组和脑缺血—亚低温组,利用大脑中动脉线栓法制作局灶性脑缺血模型,利用免疫组化及TUNEL法观察Bcl-2蛋白及神经细胞凋亡情况。结果:假手术组Bcl-2蛋白阴性表达,亦未见凋亡细胞;与脑缺血组比较,亚低温组Bcl-2蛋白表达显著增加,两组相比差异有显著性(P<0.05),神经细胞的凋亡数目明显减少,两组相比差异有统计学意义(P<0.05)。结论:亚低温可以减轻脑缺血损伤,可能与通过促进Bcl-2蛋白的表达,减少神经细胞的凋亡有关。
Objective: To study the role of mild hypothermia against neuronal apoptosis in rats with focal cerebral ischemia, Methods: Thirty clean-grade female rats were selected , and randomly divided into 3 groups: sham operation group and MCAO group and MCAO-mild hypothermia group. Model of focal ischemia was used by middle cerebral artery occlusion (MCAO). the rats were anesthetized and the brains were removed, lmmunohistochemical analysis and TUNEL staining were used to detect expression of Bcl-2 protein and neuronal apoptosis. Results: There were no positive cells of Bcl-2 protein staining and neuronal apoptosis in the sham operation group. In MCAO-mild hypothermia group, numbers of neuronal apoptosis were significantly decreased, and expression of Bcl-2 protein was significantly increased compared with MCAO group. Conclusion: Mild hypothermia may alleviate cerebral ischemic partly by increasing expression of Bcl-2 protein and decreasing apoptosis.
