辛伐他汀对大鼠肾间质成纤维细胞增殖及α_5整合素表达的影响

摘要 目的研究辛伐他汀对培养的大鼠肾间质成纤维细胞TGF-β1激活后细胞增殖及表面α5整合素表达的影响。方法不同浓度辛伐他汀加入到含有TGF-β1培养液培养的大鼠肾间质成纤维细胞,分别采用MTT法及细胞流式仪检测细胞的增殖,RT-PCR法检测大鼠肾间质成纤维细胞表面α5整合素的表达。结果辛伐他汀能抑制由TGF-β1所引起的大鼠肾间质成纤维细胞的增殖,阻止细胞由G1期进入S期。辛伐他汀能显著抑制TGF-β1对大鼠肾间质成纤维细胞表面α5整合素的表达促进作用,同时随着辛伐他汀浓度的增加,抑制作用明显增强。结论辛伐他汀能抑制由TGF-β1所引起的大鼠肾间质成纤维细胞的增殖及细胞表面α5整合素的表达。 Objective To investigate the effects of Simvastatin on proliferation and expression of α5 integrin in activated cultured rat renal interstitial fibroblasts in vitro with TGF-β1. Methods The activated cultured rat renal interstitial fibroblasts with TGF-β1 were cultured with Simvastatin by different concentrations. The proliferation and expression of a5 integrin in stimulated renal interstitial fibroblast were measured with MTT method, FCM and RT- PCR,repectively. Results Simvastatin could inhibit growth of activated rat renal interstitial fibroblasts with TGF-β1 and arrest cells in the G1 phase. Simvastatin had significantly dose-dependent stimulating inhibitory effects on TGF- β1-induced expression of α5 integrin. Conclusions Simvastatin may inhibit proliferation and expression of α5 integrin in activated rat renal interstitial fibroblasts with TGF-β1.
