摘要
目的研究人3型腺病毒(human adenovirus type 3,HAdV-3)全基因组序列的基本特征并进行基因组的进化分析。方法取患儿鼻咽分泌物进行病毒分离,用中和试验和PCR进一步鉴定分型;克隆酶切片段并测序,利用软件对腺病毒基因组蛋白进行进化树分析。结果分离得到2株HAdV-3,基因组全长分别为35273bp和35269bp,2株均含有39个DNA编码序列和2个RNA编码序列。非编码区均保守,存在回文序列和反转重复序列。进化分析表明HAdV-3及其他B组腺病毒与猿腺病毒21型具有更近的进化关系。结论获得广州地区分离的2株HAdV-3,基因组全长35273bp和35269bp;人B组腺病毒与猿腺病毒存在一定的进化关系。
Objective To obtain human adenovirus type 3(HAdV-3) field strains and the complete genome sequences and farther to have a phylngenetic analysis. Methods Nasal aspirate specimens of children were inoculated into HEp-2 and HeLa culture tubes and identified by a neutralization assay with type-specific reference rabbit antisera. Type-specific primers were also utilized to correctly identify the serotype. The restriction fragments of HAdV genome DNA were cloned into pBlueScript SK ( + ) vectors and sequenced. The 5' and 3' ends of the linear HAdV-3 genome were directly sequenced with the repurified genomic DNA as templates. General features of the HAdV-3 genome sequences were revealed using several softwares. Phylogenetic analysis were done with MEGA 3.0 soft. Results Two field strains of HAdV-3(strain Guangzhou 01 and Guangzhou 02) were isolated from children with acute respiratory infection in Guangzhou. Both genome sequences possess the same four early regions and five late regions, and have 39 DNA coding sequences and two RNA coding sequences. Other noncoding features are conserved. Inverted repeats and palindromes were identified in the genome sequences. The genomes of HAdV-B species including HAdV-3 have a strong phylngenetic relationship to chimpanzee adenovirus type 21. Conclusion Two field strains of HAdV-3 were isolated in Guangzhou, the genome length of which is 35 273 bp and 35 269 bp, respectively. The phylogenetic analysis showed that HAdV-B species have some relationship with some chimpanzee adenovims.
