rhaFGF改构体对SH-SY5Y细胞兴奋性氨基酸毒性损伤的作用 被引量:2

摘要 目的探讨rhaFGF的改构体对谷氨酸损伤神经细胞的干预作用。方法制备SH-SY5Y损伤模型,将实验分为空白组、Glu组、rhaFGF组和rhaFGF改构体组,利用流式细胞仪分析rhaFGF改构体对正常培养的神经细胞细胞周期的影响:形态观察以及MTT法检测各实验组的细胞活力、LDH法检测乳酸脱氢酶的释放率、NO法检测NO的产生。结果rhaFGF改构体组细胞S期的DNA含量为25.98%,比rhaFGF组(46.31%)显著降低(P<0.01),与空白组没有差异(P>0.05);与损伤组比较,rhaFGF改构体组(3个浓度)细胞OD值降低,LDH的释放减少,NO的过量产生减少,且均表现出一定的量效关系。结论rhaFGF改构体具有相对低的促分裂活性,仍能拮抗Glu毒性导致的细胞损伤,促进神经细胞生存,对神经细胞具有保护作用。 Objective To investigate the protective effect of Modified rhaFGF on SH-SY5Y cell impaired by excitatory anmino acids. Methods The cellular damage model in vitro was established by citatory amino acids. The effect of Modified rhaFGF on DNA synthesis was observed by Flowcytometer and the protective effect of Modified rhaFGF by different methods involved MTT, LDH and NO. Results Modified rhaFGF showed its less than mitogenic activities, and improved the survival rate of SH-SYSY and inhibited the damage of excitatory amino acids. Conclusion Modified rhaFGF could improve the survival rate of cell and protect neural cells.
