双向凝胶电泳-质谱技术筛选血清标志物方法的建立

Establishment of Screening Serum Biomarkers by Two-dimensional Polyacrylamide Gel Electrophoresis and Mass Spectrometry

摘要 目的应用双向凝胶电泳-质谱技术建立和评价血清蛋白质组学研究中白蛋白和IgG的去除方法。方法应用Proteo Extract Albumin/IgG Removal Kit纯化血清蛋白,去除白蛋白和IgG,利用双向凝胶电泳(2-DE)和质谱分析评价血清蛋白纯化效果。结果建立了稳定的血清白蛋白和IgG去除方法和双向凝胶电泳技术,且质谱证实2-DE图谱中低丰度蛋白质分辨率增加,减少了高丰度蛋白对其的干扰。结论本研究成功建立了血清蛋白质的纯化技术,为寻找疾病的血清学标志物奠定了基础。 Objective To establish and evaluate the method of removing albumin and IgG in serum proteomics by Two-dimensional electrophoresis (2-DE) and Mass Spectrometry(MS). Methods Removing serum albumin and IgG was achieved by filtering with ProteoExtract Albumin/ IgG Removal Kit and the effectiveness of purification evaluated by 2-DE and MS. Results The method of removing serum albumin and IgG and 2-DE was established stably, increasing sharp- ness of separation of low-abundance proteins and decreasing the disturbing of high abundance proteins in 2-DE which were confirmed by MS. Conclusion The established method of purifying serum proteins may be foundation in searching serology marker of diseases.
