摘要
目的:探讨祛银颗粒治疗银屑病的可能机制及量效关系。方法:建立小鼠阴道上皮模型和鼠尾鳞片表皮模型,设生理盐水组、祛银颗粒低、中、高剂量组和甲氨喋呤(methotrexatum,MTX)组,观察各组对小鼠阴道上皮细胞有丝分裂的影响和促进小鼠尾部鳞片颗粒层形成的能力。结果:祛银颗粒中剂量组及高剂量组均可以显著抑制小鼠阴道上皮细胞有丝分裂(P<0.01),不同浓度祛银颗粒呈现明显的量效关系;高剂量组与甲氨喋呤组抑制小鼠阴道上皮细胞有丝分裂效果相当。祛银颗粒中剂量组、高剂量组可以显著促进小鼠尾部鳞片表皮的颗粒层形成(P<0.01),不同浓度祛银颗粒呈现明显的量效关系;表明祛银颗粒能够促进小鼠尾部鳞片颗粒层形成,并随剂量的增大而作用增强,但促进小鼠尾部鳞片颗粒层形成的能力均不如甲氨喋呤组。研究未发现中药组有明显的毒副作用。结论:祛银颗粒治疗银屑病的机制与抑制表皮细胞增生和改善角化不全有关。
Objective： To discuss the possible mechanism and the dose-effect relationship of Quyin Granule in treating psoriarsis. Methods： The models of proliferative vaginal epithelial cells of mouse and mouse tail epidermal squama cells were set up. They were divided into five groups： the normal saline negative control group, the Quyin Granule low, middle and high dose groups and the Methotrexatum （MTX） group. The mitosis state of mouse vaginal epithelial cells and the ability of promoting the formation of mouse tail squama epidermal granular layer were observed in every group. Results： Both of the Quyin Granule middle and high dose groups could significantly inhibit the mitosis of mouse vaginal epithelial cells （ P 〈 0. 01）, and different concentrations of Quyin Granule showed evident dose-effect relationship; High-dose group had a corresponding effect in inhibiting the mitosis of mouse vaginal epithelial cells compared with the MTX group. The middle and high Quyin Granule groups could significantly promote the formation of mouse tail squama epidermal granular layer （ P 〈 0.01）, and different concentrations of Quyin Granule had evident dose-effect relationship; The result also showed that Quyin Granule could promote the formation of mouse tail squama epidermal granular layer with a positive dose-effect relationship, but the ability was lower than that of the MTX group. The toxicity and side effect were not found in the QuYin Granule groups. Conclusion： The research indicates that the possible mechanism of the QuYin Granule is the inhibition of faster epidermal cell proliferation and improvement of the parakeratotic epidermal cells.
