彭泽贝母组织培养和高产细胞系筛选研究 被引量:4

摘要 在诱导彭泽贝母鳞茎愈伤组织基础上,研究了培养过程中培养物生长、生物碱积累规律以及不同培养基和培养方式的影响。结果表明:培养50d的培养物,增殖倍数达到最高值为5.59倍,生物碱含量在60-70d出现最大值,最佳收获期为60d;基本培养基以MS更利于彭泽贝母细胞的生长和生物碱的生产;液体培养有利于培养物的生长,而固体培养更利于细胞干物质积累。用目视法从愈伤组织中初步筛选出了2个优良细胞系:分别为淡白色系(z1系)和淡绿色系(z2系)。 Based on the calli induced with bulb of FritiUaria monatha Migo, the rules of the growth of the cultured tissues and alkaloid accumulation, as well as their influence factors such as different media and culturing mode, were studied during the cultivation process. The results showed that the culture proliferation rate reached the highest, which was 5.59 times after being cultured for 50 d, while the alkaloid content reached the peak after 60 -70 d, the optimal harvest time was on the 60th day ; MS medium more profitable for the growth of the cultured tissues and alkaloid accumulation than others; liquid culture profited the growth, while solid culture was more suitable for cell dry matter accumulation; two good cell lines , a faint white ( Z1 ) and a faint green (Z2), were selected preliminary by colour with naked eyes from its calli.
