摘要
采用ISSR分子标记技术对江西、湖南、四川3个枳壳道地产区的主流品种进行遗传多样性分析,为筛选优良品种提供参考。15个引物共扩增114个位点,其中多态性位点108个,多态位点百分率为94.73%;观测等位基因数(Na)1.9467,有效等位基因数(Ne)1.5740;Ne i’s基因多样性(He)33.15%;Shannon’s信息指数(I)0.490 7;UPGMA聚类分析可将所有样品分为4类。枳壳具有较高的遗传多样性,其遗传变异主要存在于不同的品种间;聚类结果与传统的外观形态鉴别分类具有一致性;在进行优良品种选育时,应加强不同品种之间的基因交流,以提高种质的质量。
The genetic diversity of Citrus aurantium' s main current species in Jiangxi, Hunan and Sichuan Provinces was analyzed by ISSR markers, so as to make reference for sieving good species. 114 discernible DNA fragments were produced by using 15 primers,of which 108 (PPB =94.73% ) were polymorphic loci. The observed number of alleles (Na) was 1. 946 7 ; effective number of alleles (Ne) was 1. 574 0 ; Nei' s gene diversity (He) was 33. 15% ;Shannon' s information index (I) was 0. 490 7. All the samples can be divided to four types by cluster analysis(UPGMA). Citrus aurantium L. has high levels of genetic diversity and the ge- netic variation mainly exists in different species. The result of cluster analysis was consistent with that of traditional external appearance discrimination. In order to improve the quality of germplasm , when good species are sieved, gene communication should be strengthened among different species.
