摘要
通过PCR方法人工合成带有K28信号肽的死亡素突变体基因,构建了重组表达载体pRHZ-41-k28Th1,转化到裂殖酵母中进行表达,发酵液中检测到与TH1理论分子量一致的多肽分子,微量稀释法检验表达产物具有抑菌活性,并计算了粗产物对4种供试菌的最低抑菌浓度。
To obtain thanatin mutant(TH]) with antibacterial activity for further research, THI cDNA which fused with K28 signal peptide was synthesized by PCR, and ligated to shuttle vector to construct recombinant vector pRHZ-41-KTha 1. This recombinant vector was transformed into yeast cell successfully, and the expressed product was found in the fermental liquid of recombinant yeast ferment liquid by SDS-PAGE electrophoresis. The recombinant product has the antibacterial activity.
