摘要
目的从ATCC提供的、携带有基质细胞衍生因子-1(stromal cell derived factor-1,SDF-1)cDNA的质粒pCMV-SPORT6中获取SDF-1的cDNA全长,并构建SDF-1-pIRES2-EGFP真核表达载体。方法采用双酶切、回收SDF-1cDNA片段,克隆至pIRES2-EGFP真核表达载体,得到SDF-1-pIRES2-EGFP真核表达载体。结果酶切及测序结果表明SDF-1-pIRES2-EGFP真核表达载体构建成功。结论获得SDF-1基因,构建SDF-1-pIRES2-EGFP真核表达载体,为探索SDF-1修饰人骨髓间充质干细胞(Mesenchymal stem cells,MSCs)促进人CD34+细胞归巢及增殖方面的作用奠定基础。
Objective To construct the eukaryotic expression vector of stromal cell derived factor- 1 ( SDF- 1) - pIRES2- EGFP by using full SDF-1cDNA genc obtained from the vector pCMV-SPORT6 which carries the SDF-1cDNA. Methods SDF-lcDNA was acquired from the vector pCMV-SPORT6 through the Xho I / EcoR I endonuclease, and then cloned into eukaryotic expression vector pIRES2-EGFP. The obtained recombinant vector was named SDF-I-pIRES2-EGFP. Results DNA sequencing and restriction endonnclease digestion analysis indicated that the eukaryotic expression vector SDF-I-plRES2-EGFP was constructed successfully. Conclusion The SDF-1cDNA is acquired and the SDF-1-pIRES2-EGFP is constructed successfully, which provide a good experimental basis for further study on the function of SDF-1-modified human bone marrow mesenchymal stem cells to promote the CD34^+ cells homing and proliferation.
