缺氧诱导因子-1α的表达与肾癌细胞增殖和凋亡研究

Expression of hypoxia inducible factor-1α and its relation to apoptosis and proliferation in renal cell carcinoma

摘要 目的探讨缺氧诱导因子-1α(HIF-1α)在肾癌组织中表达及其与Bcl-2和PCNA的关系,了解肿瘤增殖与凋亡的特点。方法采用免疫组织化学方法检测61例肾癌石蜡标本切片HIF-1α、Bcl-2和PC-NA的表达。结果HIF-1α在肾癌组织中阳性率为57.4%(35/61),其表达与不同临床分期、病理分级有关(P<0.05),与组织学分类无关(P>0.05)。Bcl-2在肾癌组织中阳性率为42.6%(26/61),其表达与组织学分类、临床分期、病理分级均无关(P>0.05)。HIF-1α表达与肾癌细胞增殖程度呈正相关性(r=0.489,P=0.000);与Bcl-2蛋白表达无相关性(r=-0.196,P=0.127)。结论HIF-1α表达与肾癌的增殖密切相关,与肿瘤细胞凋亡无关,可能成为判断肾细胞癌转移和预后等生物学行为的重要参考指标。 Objective: To investigate the relationship of the expression of hypoxia inducible factor - 1 α ( HIF - 1 α), Bel -2 and proliferation nuclear antigen (PCNA), in renal cell carcinoma( RCC ) and to evaluate their clinical significance. Methods: The expression of HIF - 1 et, Bcl - 2 and PCNA were tested by immunohistochemical methods in 61 cases of RCC tissues, and their clinical significance were analyzed. Results :The positive rate of HIF - 1α expression in the RCC tissues was 57.4%. The expression of HIF - 1α in RCC tissues which was not related to the histological type of RCC ( P 〉 0.05 ) but related to and increased with the clinical stages and pathological grades of RCC (P 〈 0.05 ). The positive rate of Bcl -2 expression in the RCC tissues was 42.6% (26/61). The expression of Bcl - 2 in RCC tissues was not related to the histological type, the clinical stages and pathological grades of RCC (P 〉 0.05 ). The expression of HIF - 1 α was positively related with the expression of PCNA ( r = 0. 489, P = 0.000 ) , but had no relationship with the expression of Bcl -2 (r = -0. 196, P =0. 127). Conclusion: Overexpression of HIF - 1α is significantly associated with the proliferation in the RCC tissues, but has no relation with the cell apoptosis of RCC. HIF - 1α may serve as important indicator to evaluate the biological of RCC such as metastasis and prognosis.
