高效液相色谱法测定柑橘中2,4-二氯苯氧乙酸残留 被引量:30

Determination of 2,4-dichlorophenoxyacetic Acid Residue in Orange Using High Performance Liquid Chromatography

摘要 建立了柑橘中2,4-二氯苯氧乙酸(2,4-D)残留的高效液相色谱(HPLC)测定方法。样品用水提取,提取液经二氯甲烷萃取、浓缩后,用丙酮溶解。丙酮溶液上硅胶固相萃取柱,用甲醇洗脱,洗脱液氮气吹干后用流动相溶解。HPLC条件:二极管矩阵检测器检测;波长:282nm;色谱柱:ODS-3 C18柱(4.6mm×250mm,5μm);流动相:甲醇-水为8:2(体积比),pH3.0。在0.2~2.0mg/kg添加范围内,回收率86.5%~96.6%,变异系数2.45%~8.35%,最低检测限为0.05mg/kg。 A high performance liquid chromatographic method was developed for detecting 2,4-dichlorophenoxyacetic acid 2,4-D in orange. 2,4-dichlorophenoxyacetic acid was extracted from orange with water, purified by passing through silica solid-phase extraction cartridge. For analysis, a ODS-3 C18 colunm (4. 6 mm × 150 mm, 5 μm) was used at 30 ℃ and fluted with methanol-water (80 : 20, V/V, pH 3.0) at flow rate of 1.0 ml/min, and the detection wavelength was 282 nm. Average recoveries of 2,4-dichlorophenoxyacetic acid from the oranges fortified at a level of 0. 2 -2. 0 mg/kg were 86. 5% -96. 6%, with the CVs of 2. 45% - 8.35%. The detection limit for 2,4-dichlorophenoxyacetic acid was 0. 05 mg/kg.
