摘要 本文综述二甲双胍(Metformin)的临床应用及研究进展。二甲双胍通过抑制糖异生和减少糖原分解来降低肝糖输出,增加周围组织的胰岛素敏感性,降低血脂,恢复胰岛素分泌,抑制肠道摄取葡萄糖,降低餐后血糖;改善胰岛素抵抗,有心血管保护作用,改善血管内皮细胞反应性,有效降低心血管疾病的发病率和死亡率。二甲双胍最常见的副作用是胃肠道反应,乳酸中毒罕见,但死亡率高。
