摘要 目的观察TGFβ1诱导的肾小管上皮细胞转分化过程中,整合素连接激酶(integrin-linked kinase,ILK)的表达及其在TEMT中所起的作用。方法以体外培养的人近端肾小管上皮细胞系为对象,分别用TGFβ1(40ng/ml)及TGFβ1(40ng/ml)加HGF(100ng/ml)刺激,免疫组化方法检测ILK、E钙黏蛋白(E-cadherin)及纤维连接蛋白(FN)的蛋白表达,并用计算机真彩色图像分析系统对其进行半定量,并用Western Blot进一步检测ILK蛋白的表达。结果TGFβ1能以时间依赖的方式诱导ILK在肾小管上皮细胞中的表达;增加的ILK能介导肾小管上皮细胞发生表型变化,即使小管上皮细胞E钙黏蛋白的表达减少,而纤维连接蛋白的表达增多,用HGF能抑制上述作用。结论ILK参与TGFβ1诱导的人近端肾小管上皮细胞向间叶细胞的转分化。 Objective To study the expression and role of integrin-linked kinase in the renal tubular epithelial to mesenchymal transition induced by TGFβ1. Methods The normal human kidney proximal tubular cell line (HKC) was cultured in the presence of TGFβ1 (40ng/ml) and TGFβ1 (40ng/ml)1 + HGF(100ng/ml). Immunohistochemistry stain and analysis system were used to semi quantitatively examine the expression of ILK, E-cadherin and fibronectin. Results TGFβ1 induced ILK expression in renal tubular epithelial cells in a time- dependent manner. Forced expression of ILK in human kidney proximal tubular epithelial cells played an important role in mediating tubular EMT induced by TGFβ1 , via suppressing E-cadherin expression and inducing fibronectin expression. Moreover, inhibition of ILK expression by HGF was associated with blockade of tubular EMT. Conclusion ILK is a critical mediator for phenotypic transformation of renal tubular epithelial cells into mesanchymal cells indued by TGFβ1.
