摘要
目的考察生远志及其皂苷与蜜远志对大鼠胃组织中相关活性物质及酶活性的影响,以从分子水平阐释其损伤胃黏膜及抑制胃肠运动的机制。方法采用考马斯亮兰显色法、硝酸还原酶法、定磷法,测定灌胃10 d大鼠胃组织Ca2+-ATP酶活力,PGE2和NO含量。结果①与空白组相比,生远志及其皂苷与蜜远志均能明显降低胃组织PGE2含量(P<0.05或P<0.01);②生远志组与空白组相比,能显著增加胃组织中NO含量(P<0.05),而皂苷组对胃组织NO含量虽有增高趋势,但与空白组比较无显著性差异(P>0.05),蜜远志对NO影响不明显;③生远志组与空白组比较,能显著降低胃组织Ca2+-ATP酶活力(P<0.05),而皂苷组和蜜远志组与生远志比较,有显著性差异(P<0.05)。结论生远志抑制胃肠运动及损伤黏膜机制与其降低胃组织PGE2含量与Ca2+-ATP酶活力,增加NO含量相关;远志皂苷能极显著降低胃组织PGE2含量,其主要呈现胃黏膜损伤不良反应;蜜远志亦能降低胃组织PGE2含量,但对Ca2+-ATP酶与NO无明显影响。
Objective To study the effects of crude Radix Polygalae , Saponins and Honey - stir - baked Radix Polygalae on the active substances and enzyme in gaster - tissue of rat so as to explain its mechanism in molecular level. Methods Using method of coomassie brilliant blue, method of nitroreductase chromatometry and the colorimetric determination of phosphorus, we detected Ca^2+ - ATPase ,PGE2 and NO in gaster - tissue of rat. Results ①Compared with the control, the group of crude Saponins and Honey - stir - baked Radix Polygalae could remarkably decrease PGE2 ( P 〈0.05 or P 〈0.01 ). ②Compared with the control, the group of crude Radix Polygalae could remarkably increase NO( P 〈 0.05). The group of Saponins has tendency of increasing NO, but there was no remarkable difference compared with the control ( P 〉 0.05 ). There was no remarkable difference between the group of Honey - stir - baked Radix Polygalae and the control. ③Compared with the control, the group of crude Radix Polygalae could remarkably decrease Ca^2+ -ATPase( P 〈 0.05). The group of Saponins has a tendency of increasing NO, but there was no remarkable difference compared with the control ( P 〉 0.05 ). The groups of Saponins and Honey - stir - baked Radix Polygalae had no remarkable differences compazing with crud Radix Polygalae. Conclusion The mechanism of crude Radix Polygalae could restain gastrointestinal motility and damage mucosa is related to decreasing PGE2 and Ca^2+ - ATPase and increasing NO. Saponins can remarkably decrease PGE2 and has a tendency of decreasing Ca^2+ - ATPase but damage in mucosa is its chief affection. Honey - stir - baked Radix Polygalae can decrease PGE2, but it has no effect on Ca^2+- ATPase and NO.
