睫状神经营养因子对星形胶质细胞细胞周期的影响

摘要 目的探讨睫状神经营养因子(CNTF)在有血清培养和无血清培养的情况下,对星形胶质细胞分化和增殖的影响。方法将纯化的星形胶质细胞分为有血清培养和无血清培养组,根据CNTF剂量不同,每组再分别分为0、2、20、200 ng/ml 4个亚组(共8个亚组)。每个亚组分别在6、12和24 h取材,应用流式细胞术检测星形胶质细胞在各细胞周期时相中的百分比。结果CNTF使无血清培养组的星形胶质细胞处于G0/G1期的细胞百分比上升,而使有血清培养组的星形胶质细胞处于G0/G1期的细胞百分比下降。结论无血清培养时CNTF可以刺激星形胶质细胞分化进入活化状态,但不刺激其增殖;有血清培养时CNTF可以协助血清中的丝裂原引起星形胶质细胞增殖。 Objective To observe the effect of ciliary neurotrophic factor (CNTF) on the cell differentiation and proliferation of astrocytes. Methods Astrocytes purified from newborn rats'cerebral cortex were cultured in normal or serum deprivation conditions. The astrocytes were divided into 8 groups according to the concentrations of CNTF (0, 2, 20 and 200 ng/ml). The samples were harvested at 6, 12 and 24 h respectively in each group. The cell cycle of the astrocytes was examined by the flow cytometry. Results Astrocytes were cultured in the presence or absence of serum. The index of G0/G1 was increased and the index of proliferation was decreased after CNTF was applied. When astrocytes were cultured in the absence of serum, CNTF accelerated the processes of the cell cycle. The index of G0/G1 was decreased and the index of proliferation increased after CNTF treatment. Conclusion CNTF induces differentiation of astrocytes cultured in the absence of serum into active state, but doesn't stimulate the proliferation of astrocytes by itself. CNTF aids the proliferation of astrocytes cultured in the presence of serum caused by mitogen.
