摘要
目的:通过生物信息学软件对易引起慢性肝炎的三种肝炎病毒基因组结构进行分析,设计肝炎病毒诊断分型芯片的寡核苷酸(O ligo)探针。方法:利用BLAST软件对乙型肝炎病毒(HBV)、丁型肝炎病毒(HDV)、丙型肝炎病毒(HCV)6个亚型的DNA及cDNA序列进行序列比对,得到有意义的特异序列;然后用生物信息学软件Array designer 3.0设计特异性高、Tm值相近、长度均一的O ligo探针。结果:获得156条60-m er探针,用于打印成DNA芯片,进行HBV、HDV的联合检测和HCV的基因分型。结论:利用BLAST检索系统和生物学软件Array desig-ner 3.0设计肝炎病毒诊断分型芯片的探针,是一种简便可行、有效的方法。
Objective: To design oligonucleotide probes for microarray detection and genotype of hepatitis virus. Methods. By analyzing the DNA or cDNA of HBV, HDV and HCV of 6 different genotypes with BLAST program, a group of specific sequences for the candidate probes was specified. Array designer 3.0 software was applied to analyze the candidates to select the probes with high specificity, identical length and similar melting temperature(Tm). Results:Altogether 156 oligonucleotide probes were designed, and deposited on oligonucleotide chips as the microarray for hepatitis virus detection and genotype. Conclusion:BLAST and Array designer 3.0 software are effective for designing the oligonucleotide probes.
