摘要
目的研制能够与黄曲霉毒素B1、B2、G1、G2有较好交叉反应的抗总黄曲霉毒素单克隆抗体。方法利用B细胞杂交瘤技术,建立分泌抗总黄曲霉毒素单克隆抗体杂交瘤细胞株。结果建立了1株稳定分泌抗总黄曲霉毒素抗体的杂交瘤细胞株,命名为2G2。该细胞株所分泌的单克隆抗体Ig亚类为IgG1,参考工作浓度为1∶1.6×106,亲和力常数为8×10-8mol/L。该抗体与黄曲霉毒素B1、B2、G1、G2的交叉反应分别为100.0%、57.5%、104.0%和19.0%,与其他真菌毒素无交叉反应。结论制备出能分泌具有高特异性和亲和力的抗总黄曲霉毒素单克隆抗体的杂交瘤细胞株。
Objective To develop hybridoma cell line excreting monoclonal antibody against total aflatoxins. Methods Hybridoma cell line excreting monoclonal antibody against total atlatoxins was produced by using B cell hybridoma technique. Results One hybfidoma cell line excreting monoclonal antibodies against total aflatoxins, coded 2G2 was obtained by fusing routine Sp2/0 cells with spleen ceils from Balb/c mice immunized with aflatoxin Bl - BSA conjugate. The monoclonal antibody produced by the hybridoma cell was tested for subtypes and designated as IgC1.The reference working concentration was 1 : 1.6×10^6, and its affinity constant was 8×10^-8mol/L.The cross reaction between aflatoxin B1, B2, C2, G2 was 100%, 57.5%, 104%, 19% respectively, and there was no cross reactions with other mycotoxins. Condusion Monoclonal antibodies against total aflatoxins that have high specificity and affinity is produced and an ELISA detection method for total aflatoxins is found.
