摘要
目的:研究胆固醇对人血管内皮细胞基因组DNA甲基化水平的影响。方法:体外培养人脐静脉血管内皮细胞ECV304,以50.00 mg/L的胆固醇作用于ECV304 48 h,提取细胞基因组DNA,用高效液相色谱法(HPLC)检测基因组DNA甲基化水平。结果:与无胆固醇的对照组比较,50.00 mg/L胆固醇使ECV304细胞中DNA甲基化水平显著下降(P<0.01)。结论:一定浓度的胆固醇能降低血管内皮细胞中DNA甲基化水平。
Objective: To investigate the influence on the methylation level of genomic DNA caused by cholesterol on the vascular endothelial cells (VECs) . Methods: Cholesterol (50.00 mg/L) was used on human umbilical endothelial cell line, ECV304, 48 h later, cell genomic DNA methylation level was detected by HPLC. Results: Compared with the untreated control group, the genomic DNA methylation level sig- nificantly decreased when cell was treated by 50.00 mg/L cholesterol. Conclusion: The specifical concentration of cholesterol could decrease the VECs genomic DNA methylation level.
