摘要
应用流行性出血热(EHF)病毒抗体及胶体金探针对EHF患者外周血细胞进行病毒抗原的定位·光镜免疫金银染色(IGSS)结果显示患者外周血淋巴细胞及血小板表面EHF病毒抗原阳性。采用LowicrylK4M低温快速包埋电镜免疫金染色(IGS)方法,发现金探针特异性定位于淋巴细胞胞质的大小空泡和液泡内(可能为病毒芽生、成熟部位增生、扩大的高尔基囊泡)以及血小板质膜、开放管道系统扩张的管腔内。结果表明EHFV能在人外周血淋巴细胞中复制成熟。血小板表面IGSS阳性物质主要为循环血中EHFV抗原与抗体形成的免疫复合物。
Localization of epidemic hemorrhagic fever (EHF) viral antigens in the peripheral blood cells of the pa-tients with EHF was studied using colloidal gold technique. Labeling was noted on the surfaces of peripheral lymphocytes and platelets by immunogold-silver staining for light microscopy. The results of immunogold taining performed on thin sections of Lowicryl K4M---embedded material revealed that colloidal gold probes specifically localized in the vacuoles and vesicles in lymphocytic cytoplasms (probably increased and extended vacuoles around Golgi complexes where EHF viruses budded and maturated) and on the surfaces and in the dilated canales of open canalicular system of platelets. The results indicate that EHFV can repicate and maturate in the human peripheral lymphocytes and IGSS positive substances on the surfaces of the platelets mainly are immune complexes composed of EHFV antigens and antibodies in the circulating blood.
