Expression of neurogiobin in rats with chronic cerebral circulation insufficiency

摘要 目的 探讨慢性脑供血不足大鼠大脑皮质和纹状体中脑红蛋白的表达以及神经节苷脂GM1对其表达的影响。方法 24只大鼠随机分为3组:假手术组、慢性脑供血不足模型组、神经节苷脂GM1干预模型组,每组各8只。模型组通过结扎大鼠双侧颈总动脉建立慢性脑供血不足动物模型,模型建立8周后,GM1干预组每天腹腔注射GM130mg/kg,持续2周,应用免疫组化方法,观察脑组织中脑红蛋白的表达。假手术组只作双侧颈总动脉分离,不行血管结扎,其余步骤同模型组。结果 脑红蛋白主要表达于神经元中,在大脑中分布广泛。假手术组大鼠皮质和纹状体中有一定数量的脑红蛋白免疫反应阳性细胞,而慢性脑供血不足模型组较假手术组显著增多,GM1干预组较模型组进一步增加。结论 慢性脑供血不足后脑红蛋白的表达有部分代偿性提高,GM1可部分提高脑红蛋白的表达。 Objective To investigate the expression of neuroglobin (Ngb) in the cerebral cortex and striatum of the rats with chronic cerebral circulation insufficiency (CCCI) and the effect of ganglioside GM1 on it. Methods Male SD rats (n=24) were randomly into sham operation group, model group and GM1 intervention group (n=8 per group). The rat model of CCCI was established by ligating the bilateral common carotid arteries. At week 8 after modeling, GM1 was injected intraperitoneally at 30 mg/kg each day, for 2 weeks in GM1 intervention group. Expression of Ngb in the brain tissues was detected by immunohistochemistry at week 10 after CCCI. Results Ngb was mainly expressed in neurons. Ngb-positive cells were seen in many regions of the brain tissues. A few of Ngb-positive cells could be found in cortex and striatum in the sham operation group. Ngb-positive cells in the model group were more than those in the sham operation group, but less than those in the GM1 intervention group. Conclusion CCCI can increase the expression of Ngb in rat brain. GM1 may partially increase the expression of Ngb.
