摘要
本文建立了精子表面凝集素受体的胶体金标记扫描电镜术(G—SEM)并从方法学上作了探讨。得出结论:1)18nm胶体金用SEM二次电子检测,对于精子这类表面较平滑的游离细胞是可行的;2)对精子而言,G—SEM技术中样品薄层镀金不仅可行而且是必要的;3)G—SEM技术可对精子表面受体、抗原进行定位、定性、定量三结合的研究;4)本文所建立的方法简易可行,可推广应用于对精子表面其它受体、抗原的检测。作者提出了精子G—SEM的二般程序。
A simple procedure of colloidal gold label scanning electron microscopy ~r detecting the lectin receptors
of sperm surface was reported here. The results showed: 1. The 18nm particle of gold is suitable for SEM.2. In sperm, the metal coating is acceptable and necessary for distinguishing gold marker from cell surface and protecting sperm from electric damage. 3. The technique could afford a combined investigation of localization, characterization and quantification. 4. The method is sipmle and might be general.
