摘要
用铈作为酶反应捕捉物同时结合细胞增透剂Triton X—100和细胞成分稳定剂(皂角苷,Saponin),能够成功地定位出硬骨鱼精巢组织块中细胞内的酸性磷酸酶。本文的结果表明,铈/细胞增透剂方法比传统的铅方法具有特异性高、扩散少、反应充分等多方面优点。
Glutaraldehyde fixed tissue block(< lmm) of teleost testis were permeabilized with de-tergent Triton X-100 (0.0002%) and stabilized with saponin (0.002%) during the cytochemical reaction based cerium. The results in this study have shown that this mathod is a successful and efficient method to locate phosphatases of intracelluar sites in no-sliced tissue blocks.Traditional. lead method and periodic / thiocarbohydrazide / silver proteinate cytochemical stain technique also have been used to the same materials. The comparision of cerium/cell permeabilization and routine lead method has been disscussed in the paper. It can be clearly seen that the use of cerium as a cytochemJcal probe for phosphatases has many advantages over lead technique.
