摘要 二乙基二硫代氨基甲酸钠(DDTC)能明显抑制顺铂诱导鼠伤寒沙门氏菌TA100,TA98回变,有S9代谢活化系统时抑制作用增强,160.74μg/plateDDTC对0.25~2.00μg/plate顺铂抑制率为87.4%~95.7%(TA100)和89.4%~105.5%(TA98).小鼠ip顺铂1.5mg·kg-1,24h重复ip1次,每次给顺铂后1hipDDTC400~800mg·kg-1或同时igDDTC750~1500mg·kg-1.可显著降低顺铂诱发小鼠骨髓多染红细胞微核的形成. Effect of sodium diethyldi thiocarbamate (DDTC) on the mutagenicity of cisplatin was studied by Ames test and mouse bone marrow micronucleus test. DDTC had an inhibitory effect on mutation induced by cisplatin in the indicator system of Salmonella typhimurium . When the mutation of TA100 and TA98 was induced with 0.25 2.00 μg per plate of cisplatin, the inhibitory effect increased noticeably with metabolic activation. The inhibitory rate of DDTC at 160.74 μg per plate was 87.4% 95.7% (TA100) and 89.4% 105.5% (TA98). The micronucleus formation induced by cisplatin was effectively inhibited by DDTC 400 800 mg·kg -1 ip 1 h after or 750 1500 mg·kg -1 ig simultaneously with ip cisplatin 1.5 mg·kg -1 (twice at 24 h interval). The results suggest that DDTC possesses the ability to reduce the mutagenic action of cisplatin.
