摘要
采用四氯化碳(CCl_4)制成原代培养大鼠肝细胞损伤模型,观察了绞股蓝总皂甙(GPS)对肝细胞损伤的保护作用。结果表明,预先用GPS处理肝细胞1hr,可明显减轻CCl_4引起的肝细胞DNA合成速率降低,减少肝细胞ALT逸出,抑制培养上清液中MDA含量的升高和SOD/MDA比值的缩小,并与GPS浓度呈依赖关系。提示GPS能减轻CCL_4能减轻CCl_4对离体培养的大鼠肝细胞损伤,保护肝细胞DNA合成,其作用机理与抗脂质过氧化有关。
This in vitro study on the effect of Gypenosides(GPS) on carbon tetrachloride(CCl_4) induced injury in primary cultured rat hepatocytes showed that the decrease of their DNA synthesis rate and ALT leakage caused by CCl_4 were inhibited dose dependently when hepatocytes were pretreated with GPS,and the concentration of MDA was less and SOD/MDA more,in their medium,than that of injured control.It suggests the protection,which is associated with anti-lipid peroxidation,of GPS on CCl_4 induced hepatocellular injury in vitro.
