高压氧对大鼠肢体缺血再灌注所致回肠损伤的作用研究 被引量:3

摘要 目的探讨高压氧(HBO)对大鼠肢体缺血再灌注(I/R)致回肠损伤的作用及机制。方法复制I 2 h/R l h模型,分为3组,即A组(假手术组)、B组(I/R模型组)、C组(HBO治疗组)。将回肠组织行电镜病理检查,记录血清CD31、CD61平均荧光强度和CD62P的阳性率。结果与B组比较,C组回肠组织病理改变在一定程度上有所缓解。与A组比较,B组CD31、CD61及CD62P均有升高,其中CD31和CD62P升高显著(P<0.05);与B组比较,C组CD31、CD61及CD62P均有下降趋势,其中CD31和CD62P下降尤为显著(P<0.05)。结论早期行HBO治疗可显著减少急性下肢L/R大鼠模型血小板膜糖蛋白CD31和CD62P的表达,从而有助于减轻下肢I/R继发回肠损伤。 Objective To investigate the potential protective effect and mechanism of HBO on intestinal injury which is associated with hind limb isehemia/reporfusion in rat model. Methods Adult male SD rats weighing 220 -250 g were used in the study. Bilateral hind limb isehemia was induced by applying an atraurnatie ehp at the end of abdominal aorta adjacent to junction site of both femoral artery. At the end of the isehemie period, reporfusion of the limbs was achieved by releasing the clamp. The experimental rats were randomly divided into three groups, each group.contain 10 rats. (1) In sham surgical group; (2) In ischemia/ reporfusion (I/R) group; (3) In hyperbaric oxygen (HBO) group. Ultrastructural histological pathological change in small bowel was studied. The mean immunofluorescence intensity of CD31, CD61 or Positive percentage of CD62P were measured by the flow cytometry. Results In I/R group, extensive ultrastructural histological pathological injury of small bowel occurred. The ultrastructural histological pathological changes in HBO group were dearly alleviated. Compared with sham surgical group, the expression of platelet membrane glycoprotein CD31 ,CD62P in I/R group was significantly induced (P 〈 0.05). The expression of CD31, CD62P in HBO group was much lower than that in I/R group (P 〈 0.05). Conclusions Early HBO treatment can efficiently alleviate the pathological changes in intestinal dysfunction caused by rat hind limb ischemia/ reporfusion model.
