+Gz暴露对大鼠脑海马热休克蛋白70 mRNA表达的影响 被引量:1

EFFECT OF +Gz EXPOSURE ON THE EXPRESSION OF HSP70 mRNA IN RAT HIPPOCAMPUS

摘要 目的观察不同+Gz暴露对大鼠脑海马HSP70 mRNA表达水平的影响,探讨+Gz暴露致脑损伤的机制。方法100只SD大鼠随机分为对照组,+2 Gz暴露组,+6 Gz暴露组,+10Gz暴露组。各组大鼠在G暴露后的5 h1、0 h1、d、2 d4、d处死,采用原位分子杂交方法观察大鼠脑海马部位HSP70 mRNA表达水平。结果+2 Gz、+6 Gz、+10 Gz 3种水平+Gz暴露组在暴露后5 h,HSP70 mRNA表达水平均开始增加,3种水平+Gz暴露组阳性细胞数分别为27.6±4.0、51.4±6.0和14.1±2.7,均显著高于对照组(1.2±1.6,P<0.01);在10 h时达峰值,3种水平+Gz暴露组阳性细胞数分别为33.8±4.3、69.4±7.9和20.8±4.7,均显著高于对照组(2.2±2.3,P<0.01);4 d时基本恢复正常,3种水平+Gz暴露组阳性细胞数分别为1.0±1.2、5.6±3.2和0.6±0.9。在同一时间点上,+6 Gz组HSP70 mRNA表达水平最高,10 h时的阳性细胞数为69.4±7.9;+10 Gz组最低,10 h时的阳性细胞数为20.8±4.7。结论+Gz暴露对大鼠脑海马部位HSP70 mRNA表达水平有显著的影响。 Objective To observe the changes of expression of HSP70 mRNA in rat hippocampus after exposure to + Gzof different levels and to explore the manchanism of brain injury induced by + Gzexposure. Methods One hundred SD rats were randomly divided into 4 groups: control group, + 2 Gz group, + 6 Gz group, + 10 Gz group. The rats were killed 5 h, 10 h,1 d,2 d,4 d after + Gzexposure and brain specimens were obtained. The changes of HSP70 mRNA expression were measured with in situ hybridization technique. Results The HSP70 mRNA expression in three + Gzexposure groups began to rise 5 h after exposure. The number of positive neurons in three + Gzgroups (27.6 ± 4.0,51.4 ± 6.0,14.1 ± 2.7) were greater than that in control group (1.2 ± 1.6, P 〈 0.01 ) at 5 h, reached to peak [(33.8 ±4.3, 69.4±7.9, 20.8 ±4.7)vs (2.2 ± 2.3),P 〈0.011 at 10 h,and restored to normal(1.0± 1.2,5.6± 3.2,0.6 ± 0.9)4 d after exposure. The expression level was the highest in + 6 Gz group( positive neuron number: 69.4 ±7.9)and the lowest in + 10 Gz group(20.8 ± 4.7)at the same time point(10 h). Conclusion + Gzexposure can induce obvious changes of HSP70 mRNA expression of rat hippocampus.
