摘要
目的测定氨基肽酶中的一种酶(亮氨酸氨基肽酶,LAP)作为临床肝胆疾病的诊断指标并探讨氨基肽酶的临床意义,以便于系统探讨此类酶的临床应用价值。方法以亮氨酰对硝基苯胺为底物,在405 nm下进行连续监测并对健康体检者及患者标本进行测定。结果线性范围至少为0-180 U.L-1,批内不精密度为2.9%,批间不精密度为5.6%;适用于各种自动生化分析仪。测定亮氨酰氨基肽酶的最适条件:选用浓度为125 mmol.L-1的Tris-Hcl缓冲液,最适pH为7.2,温度取37℃,得出在此实验条件下的米氏常数Km为0.15 mmol.L-1,亮氨酰对硝基苯胺浓度取6.0 mmol.L-1,取健康体检者血清157例(男81例,女76例),测得x—±s为35.7 U.L-1+8.2 U.L-1,建立了适合于临床应用及自动生化分析仪的方法和参考值。结论本法是检测肝胆疾病的可靠、稳定、快速的方法之一,对肝胆疾病的早期诊断和治疗起一定的作用;氨基肽酶可以作为多种疾病的临床诊断和监测指标。
Objective Aminopeptidease is a proteolytie enzyme, leueine aminopeptideas(LAP) is one kind of aminopeptidease, leueine aminopeptideas used as an assistant diagnosis for diseases in liver and gallbladder and explore the elirtieal significance and clinical value of aminopeptidease.Methods The method presented here uses leucylparanitroanilinum as the substrate, and can continuously monitor in 405 nm, detected serum samples of health people and patient. Results It gives out report:linearity range was at least 0- 180 U· L^-1, the intra and interassay were 2.9% and 5.6% respectively. This method is suitable to the various automatic biochemistry analyzers. Here we found the optimum situation to determine LAP:the concentration of Tris-HCl is 125 retool. L^-1 and the optimum pH is 7.2, temperature is 37 ℃, under this condition, Michaelis constant (Km ) is 0.15 mmol· L^-1, ground substance concentration is 6.0 mmol·L^-1 ,with 157 health people serum samples (81 from male,76 from female),^- x ± s= 35.7 U·L^-1 ± 8. 2 U· L^- 1,we established the method and reference value for clinical application and automatic analysis. Conclusion The operation is a concise, stable and speediness method to detect diseases in liver and gallbladder, It will be a method to help diagnosing diseases in liver and gallbladder; Aminopeptidease are useful index for the diagnosis and treatment of different disease.
