色素上皮衍生因子对新生小鼠视网膜病变新生血管的抑制作用 被引量:4

Effects of pigment epithelium-derived factor on neovascularization in oxygen-induced retinopathy

摘要 目的探讨色素上皮衍生因子(pigment epithelium-derived factor,PEDF)对高氧诱导的新生小鼠视网膜病变新生血管的抑制作用。方法选取7日龄C57BL/6J新生小鼠吸75%高氧5d后返回空气制备早产儿视网膜病(retinopathy of prematurity,ROP)的小鼠模型。通过对模型组(n=26)和模型对照组(n=26)小鼠视网膜比较,观察吸氧对视网膜血管的影响。治疗组(n=112):采用不同剂量和注药次数给ROP小鼠右眼玻璃体内注入PEDF,左眼作为自身对照,在生后17d各组取材做视网膜铺片和切片,对视网膜新生血管进行形态学观察和定量研究。结果ROP小鼠在生后第12天和14天右眼注入两次PEDF后,第17天时计数突破视网膜内界膜的细胞核数,1μg组左右眼细胞核数分别是(39.43±3.28)个和(13.51±2.05)个(P=0.000);2μg组左右眼细胞核数分别是(48.40±6.24)个和(11.80±1.74)个(P=0.000)。血管形态和定量结果均显示眼内注射PEDF后新生血管形成明显减少。以2μg作为注射PEDF剂量,单次注药无效,二次和三次眼内注药有效果。结论玻璃体内注入PEDF能有效抑制视网膜新生血管的生长,同时不影响正常血管的发育,可能成为防治ROP有效的血管抑制因子。 Objective To investigate the effect of pigment epithelium-derived factor (PEDF) on inhibiting retinal neovascularization in a murine model of oxygen-induced retinopathy. Methods 7-day-old C57BL/6 mice were exposed to 75% oxygen for 5 days and then to normal situation to produce the murine ROP model. Mice in treatment group (n= 112) were received intravitreal injections of PEDF in right eyes, while left eyes served as control. The proliferative neovascular response was estimated by observing the vascular pattern in adenosine diphosphatase (ADPase) stained retina flatmounts and quantitative analyzed by counting the number of new vascular cell nuclei extending into the internal limiting membrane in cross-sections, Results There were significant differences after 1μg (left eyes 39.43±3.28 vs, right eyes 13.51±2.05) and 2 μg (left eyes 48.40±6.24 vs, right eyes 11.80±1. 74) PEDF were injected (P=0. 000), There was no effect on retinal vessels when single dose of 2 μg PEDF was injected at the time point of 12 day after born (P12) or P14, while there were responses when two or three times of injections of 2μg PEDF were injected, and there was no statistical difference between two or three times of injections (P〉0.05). Conclusions Intravitreal injection of PEDF could inhibit retinal neovascularization without overt harm to retinal morphology, It suggests that localized administration of PEDF may be an effective approach for the treatment of ischemia-induced retinal neovascular disorders such as retinopathy of prematurity (ROP).
