摘要
研究了多种因素对高羊茅胚性愈伤组织分化的影响。结果表明,BAP在高羊茅胚性愈伤组织分化中的作用要比KT大,但两者配合使用效果更佳,其适宜组合为2.0 mg/L BAP+0.5 mg/L KT。在该细胞分裂素水平下,生长素NAA宜用0.5 mg/L。胚性愈伤组织先在蔗糖浓度较高的分化培养基上预分化,以及经高渗预分化后的致密愈伤组织在琼脂浓度较高的分化培养基上分化,都能较好地促进愈伤组织植株再生,蔗糖和琼脂的适宜浓度分别为60和10 g/L。分化培养基中添加脯氨酸导致总的愈伤组织再生率降低,但同时有减少白化苗再生的趋势。
The influenees of multiple factors on differentiation of tail fescue embryogenic calli were investigated. The results showed that cytokinin BAP was more effective than KT for plant regeneration of embryogenic calli, but joint application of them produced a little better effects with optimal combination being 2 mg/L BAP + 0.5 mg/L KT. At this cytokinin level, the appropriate concentration of auxin NAA was 0. 5 mg/L. Pre-differentiation of embryogenic calli on medium containing high concentrations of sucrose and differentiation of high-osmoticum pre-differentiated compact calli on medium solidified with high concentrations of agar obviously promoted plant regeneration, the suitable concentration for sucrose and agar being 60 g/L and 10 g/L , resectively. Supplementation of L-proline in differentiation medium led to decrease in frequency of regenerated callus, but simultaneously displayed a tendency to reduce regeneration of albino plantlets.
