摘要
目的观察维拉帕米缓释剂对实验性血管内膜损伤后动脉粥样硬化进程及血管平滑肌细胞(VSMC)增生的影响。方法选雄性日本大耳白兔27只,随机分为3组,A组始终给予正常饮食;B,C组予高脂喂养并在1周后,经球囊造成右颈总动脉内膜损伤。C组给予维拉帕米干预。所有组均于内膜损伤后2周各取2只处死,其余于8周后处死,于实验结束时测量内膜厚度及内膜中膜厚度比。并分别在实验开始前、实验开始后第3周及第9周抽取静脉血测量血脂含量,血清一氧化氮(NO)和血浆内皮素(ET)浓度。结果9周末B组动脉内膜较C组明显增厚(P<0.01),内膜中膜厚度比分别为(3.01±0.26)和(2.02±0.09)。B与C组较A组血中NO降低,ET-1升高(P<0.05),其变化幅度B组明显大于C组(P<0.05)。B与C血脂较A组均升高(P<0.01),但B和C之间未见差异无显著性(P>0.05)。结论维拉帕米缓释剂可通过降低ET,升高NO,抑制平滑肌细胞增殖,减轻粥样斑块形成。
[Objective] To observe the effect of Verapamil retard tablets on the development in atherosclerosis of rabbits and the proliferation of vascular smooth muscle cell. after transluminal angioplasty. [Method] 27 male rabbits were divided into 3 groups. Group A received normal chow, group B received high hpid diet. group C received high lipid diet plus verapamil retard tablets 0.12 g/(kg·d). All rabbits were fed 9 weeks. At the beginning of 2nd week, group B and group C underwent the injury of right common carotid artery with balloon. At the 3rd week, every group was randomly chosen 2 rabbits to kill. And at the end of study, the remnant rabbits were all killed. The thickness of intima was measure by image analysis, Before the study, at the end of the 3rd week and the 9th week all rabbits were collected blood sample to measure the concentration of serum cholesterol, nitric oxide, plasma endothelin. [Result] At the end of study, the intima of group B were thicker than group C (P 〈0.01). The ratio of intima to media was(3.01±0.26) (group B),( 2.02±0.09)(group C). The concentration of serum nitric oxide in group B, group C were lower than group A. But The concentration of plasma endothelin were higher (P 〈0.05). The amplitude of chang in group B exceeded group C (P 〈0.05). The level of serum cholesterol in group B ,group C were also higher than that of group A (P 〈0.01), but no difference was detected between group B and group C (P 〉0.05). [Conclusion] Verapamil retard tablets can inhabit the proliferation of vascular muscle cell and delay the development of atherosclerosis through decreasing the level of plasma endothelin and augmenting the level of serum nitric oxide.
