摘要
目的观察赶山鞭中四个黄酮化合物对PC12细胞DNA氧化应激损伤的防护作用,探讨赶山鞭防治心衰的机理和有效成分。方法体外细胞培养,H2O2处理PC12细胞复制氧化损伤模型。MTT测定细胞存活率,单细胞电泳分析DNA损伤。结果与对照组相比,四个黄酮大剂量组的细胞存活率升高(P<0.05,P<0.01),其作用强度依次为槲皮素>槲皮素-3’-O-L-鼠李糖苷>槲皮素-4’-O-β-D-葡萄糖苷>槲皮素-3’-O-β-D-葡萄糖苷。慧星出现率降低(P<0.01)。结论四种化合物均对H2O2导致的细胞氧化损伤有防护作用。
Objective To observe the effect of flavonoids from hypericum attemuatum on relieving oxidative stress injury of PC12 cell DNA. Method PC12 cell was exposed to H2O2 in vitro to establish oxidative injury model. MTT staining was applied to analyze the survival rate of cell and DNA injury was detected by single cell gel eleetrophorisi. Results The cell survival rates of all the four flavonoids large-dose groups were higher than H2O2 group (P〈0.01,P〈0.05, queroetin〉queroetin-3' -O- L-rhamnoside〉quercetin-4' -O -β-D-glueoside〉queretin- 3' -O- β-D-glucoside), while the incidence of comets was lower than H2O2 group (P〈0.01). Conclusion All of these four kinds of flavonoid had protective effects on oxidative injury cell induced by H2O2.
