摘要
采用5'加端PCR从K562cDNA文库中扩增出含锌指结构域的cDNA基因片段,重组至pGEM7ZDNA载体中。通过PCR及Southern印迹杂交初步从K562cDNA文库中筛选出9个含锌指基因片段的重组克隆。经DNA序列分析和基因库检索,发现有2个克隆的序列与巳知基因差异显著,有可能为新的锌指蛋白基因片段。
he fragments of cDNA gene coding for zinc-finger proteins were amplified by add-on PCRfrom K562 cells cDNA library.After being cloned into pGEM 7Z Vector,nine of the inserts con-taining zinc-finger construction were identified by PCR and Southern blotting.Two of the recom-binant fragments were found to have many differences in sequences from the known genes by se-quencing and searching for the sequence data in Gene bank.These two recombinant fragmentsmay be cDNA gene fragments coding for new zinc-finger protein。
