用电穿孔技术向Ψ-2细胞导入EBV Lmp基因的研究 被引量:1

摘要 采用电穿孔基因转移技术向Ψ-2细胞内导入EBVLmp一neo基因,通过G418筛选获得86个抗性克隆。用PCR检测进一步确定了外源基因在抗C418克隆中稳定的整合。研究了部分电学参数对细胞存活及基因转移效率的影响以及电穿孔基因条件的优化。 his paper reported the introduction of pMV/LMP/C-myc-neo fusion gene into Ψ-2 cells byelectroporation. The 86 resistant colonies were obtained by selective medium which containedG4l8 sulfate;the stable integration of foreign gene(LMP)in Ψ-2 cells were determined by PCR.Several parameters of electroporation were investigated.The optimal electroporation conditionsand optimal efficiency of gene transfer were described.In summary,electroporation is a usefulmethod to obtain transient or stable expression of foreign genes in animal and plant cells.
