摘要
目的建立立体定向手术下6-OHDA黑质毁损的帕金森病恒河猴模型并建立客观评估体系。方法实验用恒河猴5只,在立体定向技术下以6-OHDA对恒河猴右侧黑质进行7个靶点的毁损,术后2周、2、4、6个月进行行为学评分量化,于术后7个月以SPECT检测恒河猴双侧纹状体DAT的代谢来判断毁损侧DA代谢减少程度,术后8个月以TH免疫荧光法检测恒河猴双侧黑质的DA能神经元改变,以此来证实黑质的毁损程度。结果术后7个月SPECT检测显示毁损侧纹状体区DA代谢仅为对侧的10%~25%左右,术后8个月双侧黑质的TH免疫荧光检测显示毁损侧黑质DA能神经元减少70%以上。结论立体定向技术下6-OHDA单侧恒河猴黑质毁损方法能够成功的制作符合人类病理特征的帕金森病模型。SPECT是评价模型制作成功与否的客观手段。
Objective To establish a stereotactic unilateral substantia nigra lesion of monkey model of Parkinson' s disease by 6-OHDA and to create a system on the assessment of the model. Methods Five adult rhesus monkeys were prepared in the study,10μl of 6 OHDA injected stereotaxicaUy into 7 targets in the right substantia nigra in every monkey respectively,then 2 weeks,2 months,4 months and 6 months later all monkeys received the behavioral test, 7 months later all monkeys received the DAT checking by SPECT using 99mTc-TRODAT-1 ,and 8 months later,TH-positive cells were detected in the bilateral substantia nigra by immunoflurescence methods. Results The examination of SPECT showed that the dopamine metabolism was decreased by about 75 % in the right striatum compared with the intact striatum. The TH-positive neurons were decreased by about 70% in the right nigra compared with the left nigra. Conclusion The pathologic character of the lesions in the monkey model was approximated to the clinic Parkinson' s disease. The SPECT can provide an exact assessment to evaluate the Parkinson's disease monkey models.
