莪术油对中波紫外线辐射致豚鼠皮肤损伤的治疗作用 被引量:10

摘要 目的:研究莪术油对中波紫外线(ultraviolet B,UVB)所致豚鼠皮肤损伤的治疗作用及其机制。方法:UVB照射豚鼠皮肤,建立氧化损伤模型(照射30 d,累积28.38 J.cm-2)。实验观察皮肤红斑形成、粗糙程度等外观变化及HE常规染色后光镜下表皮厚度和真皮中成纤维细胞数量的变化。酶法检测皮肤匀浆上清液中过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量及总抗氧化能力(T-AOC)。结果:①光镜下可见UVB照射组和基质空白组照射侧表皮增厚,成纤维细胞数量减少,而未照射侧未见此病理变化;莪术油治疗组照射侧未见表皮增厚,而成纤维细胞增多;但未照射侧未见此病理变化。②与正常组比较,UVB照射组和基质空白组照射侧皮肤中MDA含量明显增多,CAT、GSH-Px和SOD活性及T-AOC降低;莪术油治疗组照射侧MDA减少,CAT、GSH-Px、SOD活性及T-AOC增加;各组未照射侧无明显改变。结论:莪术油具有对UVB皮肤氧化损伤的治疗作用,其作用机制可能与提高抗氧化酶含量及清除体内自由基有关。 Objective To study the therapeutic effects of oleum curcumae wenchowensis on skin of guinea pigs damage irradiated by UVB and its mechanism. Methods The skin of guinea pigs was irradiated by UVB (28.38 J · cm^- 2,30 d) to establish the oxidative damage model. The skin erythema and the rough level were observed during the experiment; the appearance and thickness of epiderm and the number of fibroblast were observed under light microscope. The activities of GSH-Px, SOD, CAT and T-AOC and the content of MDA in the supernate of skin homogenate were detected with biochemical methods. Results The epiderm in UVB irradiated group and blank group thickened, the number of fibroblasts decreased; and the epiderm in therapeutic group didn't thicken, but the number of fibroblasts increased, but those in the unirradiated side of every group did not change. Compared with normal group, the contents of MDA in the supernate of skin homogenate in UVB irradiated group and blank group increased, but that in therapeutic group decreased. The activities of GSH-Px, SOD, CAT and T AOC in UVP, irradiated group and blank group decreased, but those in therapeutic group increased, and those in the unirradiated side of every group didn't change. Conclusion Oleum curcumae wenchowensis has the therapeutic effects on skin damage irradiated by UVB, which may be mediated by increasing the contents of antioxidases and eliminating the free radical.
