摘要
采用地高辛标记生长抑素反意RNA探针经原位杂交和显色后,光学显微镜下观察生长抑素mRNA在大鼠脊髓内的定位。结果显示:脊髓内含有大量呈紫蓝色的生长抑素mRNA阳性神经细胞,碱性磷酸酶反应产物定位于胞浆内,胞核不着色,生长抑素mRNA阳性胞体主要分布于脊髓背角Ⅱ层,Ⅲ Ⅳ、V和X层内有少量生长抑素mRNA阳性胞体,Ⅶ和Ⅷ层内仅偶见生长抑素mRNA阳性胞体。
In situ hybridization of somatostatin mRNA in the rat spinal cord was observed at light microscopic level with digoxengenin-labelled somatostatin antisense RNA as the probe. We visualized numerous purple-blue somatostatin mRNA-positive neurons in the spinal cord. Alkaline phosphatase reaction products were concentrated in the cell cytoplasm. The nucleus was unlabelled. A lot of somatostatin mRNA-positive neurons were found in layer Ⅱ and a few in layers Ⅲ, Ⅳ, V, X and occasional in layers Ⅶ,Ⅷ.
