慢性疼痛是一个世界性难题,其治疗效果不佳。与其目前机制不明有很大美系,解决机制问题,发展有效的治疗方法已经或为一个普遍的主题,RNA干扰就是一种有潜力的新方法,通过输入siRNA可选择性降解疼痛相美蛋白的mRNA,从而解除疼痛。而且作为新的“基国敲除”手段.可用于发现疼痛通路中的新基因,慢性疼痛通路中某些兴奋性异常增高的位点因而成为siRNA的干扰目标。
Chronic pain is a major socio-economic burden, mechanism of which remains unknown. It is now focused on searching new explain of chronic pare and developing more efficacious therapies. A recent development involving RNA interference (RNAi) in a pathophyaiaiogicat model of chronic pain highlighted a novel approach to potentiai treatment in humans. Delivery of bligonucleofides led to a pofeof and highly selective degradation of a target mRNA and produced a measurable abrogation in pain-relatad behaviour. It can also be used te search for new possible gene in chronic pain pathway. Tbls review will discuss the potential or small interfering RNA (siRNA) in the treatment of chronic pain by evaluating points of intervenden in chronic pain pathways that may be amenable to siRNA application.
