关木通浸膏加低盐饮食建立大鼠慢性马兜铃酸肾病模型 被引量:1

摘要 目的探讨用关木通浸膏加低盐饮食建立大鼠慢性马兜铃酸肾病动物模型的方法。方法制备关木通浸膏剂,并测定其中马兜铃酸(AA)含量。实验分为正常对照组、低剂量组(AA5mg·kg-1·d-1)、中剂量组(AA10mg·kg-1·d-1)、高剂量组(AA20mg·kg-1·d-1)。给药前一次性腹腔注射速尿4mg/kg,然后分别灌胃不同浓度的关木通浸膏,给药组给予低盐饮食(钠含量<0.05%);对照组灌胃同体积自来水。连续给药1周,停药1周,隔周给药。各组每周测量体重,每2周代谢笼法留取24h尿液;每组分别于第10d、30d处死大鼠各2只,第56d处死其余大鼠,腹主动脉采血,测定相关肾功能指标;取肾组织进行形态学检查。结果AA用药组尿量均明显增多,体重明显降低,尿蛋白、尿酶排泄量、肌酐等均升高。肾脏病理出现明显的小管间质损伤和典型的早期间质纤维化表现。结论表明成功建立了大鼠慢性AAN模型,且本方法造价低廉,造模时间短,成功率高。 Objective: To research the method of using Caulis Aristolochiae Manshuriensis(CAM) Extracta plus low salt food to make a rat model of Aristolochic Acid nephropathy(AAN), Methods:36 Wistar male rat were randomly divided into 4 groups. Every experimental groups had 10 rats, firstly received a single dose of furosemiede(4 mg·kg^-1 · d^- 1 ), then administered CAM Extracta intra - gastically, respectively by low dosage(AA 5 mg·kg^- 1·d^- 1 ), middle dosage(AA 10 mg· kg^-1- · d^-1 ), and high dosage(AA 20 mg·kg^-1·d^-1) ;so as to be fed a low - salt (0. 05 % "sodium ), normal protein diet . the control group administered equal volume of potable water. The body weight were recorded weekly, Urine collection was performed every 2 weeks . 2 rats were killed at the 10 d, 30 d, at the end of 8 th, killed the other rats. The blood samples were taken from abdominal artery for detection of indexes of renal functions. The .serum creatinin, urea nitrogen, urinary protein content and the morphological change were observed. Renal tissues were taken for detection. Results: After given AA, the urine excretion increased, the body weight declined markedly, especially in the middle and high dosage groups. The urine excretion of albumin and NAG enzyme, the level of serum creatinin levels increased in the middle and high dosage. The morphological finding showed renal tubular- interstitial injury and the early fibrosis, which were more seriously in the groups of AA 20 mg·kg^-1·d^-1 This indicated a chronic Aristolochic Acid nephropathy model was established in Wistar rats. Conclusion: A rat chronic aristolochic acid nephropathy model was successfully established, This method had advantage of cost low, shorttimes, and more success.
