摘要
利用无血清培养的鸡卵泡(F3)膜细胞模型研究了催乳素(PRL)对卵泡膜细胞类固醇激素合成的作用。用不同剂量的PRL、黄体生成素(LH)及促卵泡激素(FSH)分别或共同处理在无血清培养液中培养了48h的鸡卵泡膜细胞,24h后收集培养液,测定培养液中睾酮(T)和雌二醇(E2)含量。结果表明,LH能明显刺激膜细胞合成T和E2,并具有剂量依赖关系;PRL单独对膜细胞类固醇合成无明显作用,但可抑制LH诱导的T和E2合成,以及FSH刺激的E2合成。本研究证明PRL能作用于鸡卵泡膜细胞。
The present study revealed the effect of prolactin (PRL) on steroidogenesis of the domestic hen theca cells in the serum free 199 medium supplemented with EGF, insulin (Ins) and transferrin (Tf), PRL with or without LH/FSH were added the serum free medium containing hen theca cells. The results showed that theca cells in serum free medium response to increasing dose of LH, the testosterone (T) and estradiol (E 2) content of the media significantly increased in a dose dependent manner. PRL alone had no effect on steroidogenesis of theca cells, but when add together with FSH (100 μg/L) or LH (50 μg/L) it effectively suppressed the production of E 2 or E 2 and T which were stimulated by FSH or LH. The results suggested that PRL may have a direct inhibitory effect on gonadotropin stimulated steroidogenesis of theca cells in the domestic hen.
