吲哚草酰胺衍生物YB-15对Hela细胞生长抑制的研究

摘要 [目的]研究吲哚草酰胺衍生物YB-15对Hela细胞的作用。[方法]体外培养宫颈癌细胞Hela,应用MTT法测定半数抑制浓度(IC_(50)),荧光染色观察细胞凋亡及绘制7 d内的生长曲线观察生长情况,间接免疫荧光染色观察化合物对微丝管蛋白形态的破坏情况。[结果]吲哚草酰胺衍生物YB-15能明显抑制Hela细胞的生长,IC_(50)约为1.25×10^(-6)mol/L;1×10^(-6)mol/L吲哚草酰胺衍生物YB-15作用24 h即可明显诱导Hela细胞的凋亡;[结论]吲哚草酰胺衍生物YB-15对人类宫颈癌细胞有极强生长抑制和诱导凋亡作用。 [Objective] To study the effect of indol-3-yl-glyoxglic acid amide derivative-YB-15 on Hela cells. [Methods] Hela cells were treated by indol-3-yl-glyoxglic acid amide derivative-YB-15 at different concentration in vitro. Inhibition of growth was analysed by MTT assay, cell apoptosis was observed by fluorescence microscopy, cell cycle was determined by flow cytometry and the cell growth curves in seven days were mapped by cell counting. The effects of indol-3-yl-gly- oxglic acid amide derivative-YB-15 on the microtubule cytoskeleton were investigated by fluorescence microscopy. [Results] The growth of Hela cells was inhibited remarkably by indol-3-yl-glyoxglic acid amide derivative-YB-15. The ICs0 value of indol-3-yl-glyoxglie acid amide derivative-YB-15 for Hela cell was 1.25 ×10^-6 mol/L. Apopotosis of Hela cell was induced efficiently by indol-3-yl-glyoxglic acid amide derivative-YB-15 at the concentration of 1 ×10^-6 mol/L. [Conclusion] Indol-3-yl-glyoxglic acid amide derivative-YB-15 can efficiently inhibit the growth of Hela cells and induce apoptosis.
