摘要
我们利用尼龙膜和^(35)S标记的HPV-16探针(比活性为2-3×10~7cpm/μg),采取简单易行的斑点杂交技术,建立了消除HPV各型间交叉反应的最适杂交、漂洗和放射自显影条件,并在该条件下在30例取自天津的人宫颈鳞状上皮癌组织DNA中,检出17例(56.7%)为HPV-16DNA阳性。
Accurate typing of the different HPV types is essential in view of the different pathological potential of the common virus types of HPV present in the cervix. We have developed hybridization,washing and autoradiography conditions that minimize cross-hybridization among different specific types of HPV so as to allow clear-cut type assignment through practical dot blot hybridization techni- que using nylon membrane and 35S-labeled HPV-16 DNA probe. Under these conditions seventeen of thirty ( 56.7% ) squamous cell carcinomas of the cervix uteri obtained from Tianjin women were detected the presence of HPV-16 DNA.
