星形细胞瘤原位DNA定量

摘要 应用图象分析仪,对41例星形细胞瘤进行了原位DNA定量研究。以小淋巴细胞作为正常二倍体(2.22±0.32 AU)对照,绘制DNA分布直方图。Ⅰ~Ⅱ级星形细胞瘤呈2C~4C分布模型,Ⅰ级峰值位于2c,Ⅱ级位于3C~4C,且有一定数量的多倍体细胞(28.5%)Ⅲ~Ⅳ级星形细胞瘤DNA含量明显增加(7.13±1.82 AU,6.76±0.81 AU),直方图呈弥散、多峰分布模型,峰值位于异倍体或非整倍体。研究显示,异倍体和非整倍体细胞的出现是恶性的重要标志,Ⅱ级肥胖型星形细胞瘤有较高的恶变倾向。 Forty one astrocytomas with different grades were studied by imagp analysis technique for measuring DNA contents. The small lymphocytes were used as normal control DNA diphoid value (2.22 ± 0.32 AU). The histograms of nuclear DNA contents were made. The results showed that the distribution of DNA contents in grade Ⅰ and Ⅱ astrocytomas was 2C-4C.The peak value of grade Ⅰ astrocytoma was2C;grade Ⅱ was 3C-4C with some hypertetraploid (28.5%). The DNA contents of grade Ⅲ and Ⅳ astrocytomas were much higher (7.13 ± 1.82 AU, 6.76 ± 0.81 AU). Their histograms were dispersive with many peaks which were aneuploid and heteroploid The data confirm that hcteroploid and aneuploid were the marks of malignancy. The grade Ⅱ gemistocytie astrocytoma has the tendency to higher malignancy.
