摘要
本文利用体外培养血管内皮细胞的方法,观察了酵母多糖活化的多形核白细胞(PMN)对内皮细胞的影响。本研究发现:将活化的PMN与内皮细胞孵育,10h内皮细胞出现核固缩等变化,培养液中LDH升高;24h内皮细胞变化更为明显,细胞变性坏死,培养液中LDH和6-酮-PGF_1α含量均明显升高。予先向培养液中分别加入超氧化物歧化酶(SOD)或过氧化氢酶(CAT),内皮细胞仍有损害,将SOD和CAT予先联合加入,可保护内皮细胞免受PMN的损害。据此认为:活化PMN引起的内皮细胞损伤可能主要系羟自由基(OH·)所致。
The effect of zymosan-activated polymorphomiclear neutrophils (PMN) on the cultured vascular endothelial cells in vitro is observed. After incubating with the activated PMN for 10 or 24 h. the endothelial cells are denatured and necro-tized, and the levels of LDH and 6-keto-PGF,α in the media are higher than those measured before incubation. This phenomenon indicates that the endothelial cells are injured by the activated PMN. If SOD and catalase (CAT) are added to the media before incubation for 10 or 24 h. no observed damage is found in the endothelial cells. When SOD or CAT is added singly no protective effect is observed. According to the results it is concluded that the free adicals OH may play a main role in the activated PMN-mediated endothetial injury.
