健脑胶囊防治泼尼松性大鼠神经系统损伤的作用研究

摘要 目的探讨长期大量应用泼尼松对大鼠中枢神经系统的毒性作用,同时观察中药复方健脑胶囊对泼尼松性神经系统损害的防治作用。方法4月龄SD雄性大鼠50只随机分成5组,除正常组外其余4组予泼尼松2.7 mg.kg-1灌胃,1次/d,连续12周。健脑胶囊治疗组同时予健脑胶囊3种剂量提取液(2.5,5.0,10.0 g.kg-1.d-1)予治疗。实验中测试大鼠的应激记忆力,测定脑匀浆中MAO,SOD,AchE、MDA的含量以及对大脑进行组织学检查。结果单用泼尼松大鼠的三臂电迷宫正确率、脑匀浆中的SOD含量显著下降而MAO含量上升(P<0.01);脑神经元发生水肿变性,皮质厚度变薄,皮层和海马结构紊乱,而健脑胶囊可有效保护神经元及升高SOD的含量及降低MAO的含量以及提高迷宫正确率(P<0.05)。结论泼尼松长期应用可造成大鼠中枢神经系统功能减退,健脑胶囊对其有较好的对抗作用,但量-效关系不明显。 Objective To study the cerebral damage induced by long - term administration of prednisone, and investigate the prevention effects of Jiannao capsule on rats. Methods Fifty four-month SD male rats were randomly divided into 5 groups with 10 rats per group. Group 1 was treated orally with vehicle as control ( NS group ), other groups were administered oraly with prednisone (2.7 mg·kg^-l·d^-l ) first, and then with vehicle ( PD group), or water extract of Jiannao capsule with three different doses (2.5,5.0, 10.0g · kg^-1· d^-l ), once a day for 12 weeks. Then the 10% cerebrums inhomogenate was used to test the content of mono-amine oxidase (MAO), superoxide dismutase (SOD) and acetylcholinesterase (AchE). The right half of cerebrums was used for testing the histological observation. Results Biochemical parameters of cerebrums plasm showed that the concentration of MAO increased, SOD and AchE decreased significantly in PD group. Moreover, histopathology showed that the structure of cerebrum cortex became thinner and hippocampi was in disorder. Dropsical and necrotic nerve cells were found. The water extract of Jiannao could prevent the changes of nerve centre in prednisone rats. Conclusion The treatment of Jiannao Capsule can prevent the damage of nerve centre which caused by prednisone.
