摘要
使用特异荧光探针定位术,对不同培养时期人喉癌细胞系(HEp2)MTS表层细胞的F-肌动蛋白微丝骨架进行了对比观察。结果表明,随着多细胞肿瘤球体(MTS)转移培养时间的延长,表层细胞中出现数目较多的F-肌动蛋白小体,其直径约为0.3~1.0μm。F-肌动蛋白微丝结构的改变,可能与MTS表层细胞的侵袭与转移活性增强有关,但也有可能是表层细胞在不同生理状况下的一种反应。
We have compared the cytoskeleton of F-actin microfilaments in the outer cells of multicellular tumor spheroids(HEp 2) with different culture duration by staining with TRITC labeled phalloidin. Results showed that many F-actin aggregates appeared in the outer cells with the prolongation of MTS culture. These aggregates are dot-like with diameter 0. 3~1. 0 μm. The reorganization of F-actin microfilaments may not only be related to the greater potential capacity of outer cells to invade and metastasis ,but may also reflect the physical condition of the outer cells.
