摘要
利用四极杆-飞行时间(Q—TOF)电喷雾多级串联质谱(ESI—MS/MS)与诱导碰撞解离技术(CID)在负离子模式下选择合适碰撞能量对几组互为异构体的人乳寡糖进行了分析,并对其碎裂机理进行了探讨。结果表明:在二级质谱中碰撞能量范围为15—30eV时,寡糖异构体产生其特征碎片离子,据此可以快速灵敏地区分寡糖异构体。
Electrospray ionization-collision induced dissociation-tandem mass spectrometry(ESI-CID-MS/MS) is becoming one of the most important tools for structure analysis of oligosaccharides. In this study, a combined ESI-CID-MS/MS method on quadrupole-time of flight (Q-TOF) was developed to differentiate several isomers in human milk oligosaccharides. Under MS/MS conditions, negative ion mode, changing the CID eneryg from 15 eV to 30 eV, some useful fragments were obtained and the fragmentation mechanisms were proposed. The experimental results demonstrate that isomers in human milk oligosaccharides can be distinguished by their characteristic fragment ions quickly and conveniently
